background: metronidazole, a 5-nitroimidazole derivative, is the main antitrichomonal agent of  choice   for   treatment  of t richomoniasis . since 1962, s ome cases of treatment failure with metronidazole have been reported and recently drug resistance is now on the rise in the world. this study was aimed to determine current susceptibility of iranian isolates of trichomonas vaginalis to metronidazole. methods: this study was performed on 50 t. vaginalis isolates collected from west and central areas of iran. after axenisation of the parasites, susceptibility testing was carried out by using serial twofold dilutions of metronidazole (400 to 0.1 µg/ml). the minimum inhibitory concentration (mic) and the minimum lethal concentration (mlc) of the trichomonads were determined after 48 h incubation at 35.5 °c. drug susceptibility assays of the all isolates were carried out two times in triplicate under aerobic and anaerobic conditions. results: ninety-eight percent of the t. vaginalis isolates (49/50) were sensitive to metronidazole. metronidazole resistance was defined as aerobic mic ≥50 µg/ml, detected in one isolate. the means of aerobic mics and mlcs and that of anaerobic mics of the parasites were 2.91, 1.95 and 0.28 µg/ml, respectively. conclusion: this investigation showed in vitro low-level tolerance to metronidazole in a few t. vaginalis isolates that may be leading to the development of drug resistance.

Drug Susceptibility of Trichomonas vaginalis to Metronidazole in Bushehr City (In Vitro)

Background: Trichomoniasis is the most common non-viral sexually transmitted disease caused by the protozoan parasite Trichomonas vaginalis. Metronidazole (MTZ), a 5-nitroimidazole derivative, is the main antitrichomonal agent of choice for the treatment of trichomoniasis. Since 1962, some cases of treatment failure with metronidazole have been reported, and the increased resistance of the para...

Susceptibility of Trichomonas vaginalis to metronidazole: A survey of 76 random isolates

Isolates were cultured in an TYM-medium (MEINGASSNER and HEYWORTH 1982) containing antibiotics for inhibition of bacterial and fungal growth. Inhibitors were omitted from the growth medium when the cultures proved to be free from contaminants. Drug testing on the isolates was usually performed within 10 passages after isolation. The laboratory procedure for susceptibility testing to metronidazo...

Metronidazole-resistant Trichomonas vaginalis.

A 36-year-old woman with symptomatic metronidazole-resistant trichomonal vaginitis for 10 years had a total of 22 courses of treatment with either metronidazole or tinidazole according to different schedules. The minimum trichomonicidal concentration of metronidazole for the strain of Trichomonas vaginalis isolated from the patient was 160 microgram/ml compared with 1.25-10 microgram/ml for oth...

In vitro susceptibility of Trichomonas vaginalis to 50 antimicrobial agents.

We determined the susceptibilities of five strains of Trichomonas vaginalis, one of which was metronidazole resistant, to 50 antimicrobial agents. For the metronidazole-susceptible strains, the most active agents were metronidazole, tinidazole, mebendazole, furazolidone, and anisomycin. Against the resistant strain mebendazole, furazolidone, and anisomycin were the most active. Antifungal agent...

Ferredoxin Gene Mutation in Iranian Trichomonas vaginalis Isolates

BACKGROUND Trichomonas vaginalis causes trichomoniasis and metronidazole is its chosen drug for treatment. Ferredoxin has role in electron transport and carbohydrate metabolism and the conversion of an inactive form of metronidazole (CO) to its active form (CPR). Ferredoxin gene mutations reduce gene expression and increase its resistance to metronidazole. In this study, the frequency of ferred...
